Abstract

1,132,676. Anti-tumour preparation. CHUGAI SEIYAKU K.K. and KYOWA HAKKO KOGYO CO. Ltd. 3 March, 1967 [8 March, 1966], No. 10290/67. Heading A5B. An anti-tumour preparation containing the cells of Streptococcus hemolyticus having no virulence and no hemolytic activity but having anti-tumour activity is prepared by mixing penicillin and living cells of Streptococcus hemolyticus having anti-tumour activity in a suspension medium comprising an aqueous solution of salts, the amount of penicillin being such that the concentration of penicillin in the suspension medium exceeds 25,000 units per ml. of the total liquid in said mixture, and incubating the resulting mixture firstly at a temperature in the range of 30‹C to 38‹C and then at a temperature in the range of 38‹C to 50‹C so as to obtain the desired antitumour preparation in which the majority of the Streptococcus hemolyticus cells are dead. The first stage of incubation may be effected for 10 to 30 minutes and the second for 20 to 60 minutes. The concentration of penicillin in the suspension medium may be from 27,000 to 60,000 units/ml of the total liquid in the mixture. The suspension medium may be phosphate buffered saline or Bernheimer’s basul medium composed of maltose, KH 2 PO 4 , MgSO 4 .7H 2 O, distilled water and having a pH of from 6.8 to 7.0. The cells of Streptococcus hemolyticus may be obtained by cultivating Streptococcus hemolyticus in a cultivating medium in which the main carbon source is one other than glucose e.g. meat infusion broth or a medium containing ribonucleic acid and/or ribonuclease core each present in an amount of from 0.5 to 1.0% w/v. Penicillin may be added to the suspension medium prior to or after suspension of Streptococcus hemolyticus cells therein. The resulting suspension of cocci may be aseptically diluted with a salt solution, preferably containing penicillin to obtain a form suitable for therapeutic administration e.g. injection.
