1286883 Preparing microscope specimens F GRASENICK & E JAKOPIC 24 Sept 1970 [24 Sept 1969] 45610/70 Heading G2J [Also in Division H1] An organic sample is prepared for electron microscope examination by bombardment with electrons of energy less than 10 Kev to polymerize a thin surface layer and make it less soluble, the soluble non-irradiated part being dissolved away. During irradiation the sample may be at room temperature and it is preferably disposed within an enclosure cooled to -130 C. with liquid nitrogen to prevent contamination from hydrocarbons within the vacuum chamber. The specimen may be film of polyvinylalcohol, polyethylene terephthalate or acetobutyrate, or may be a millipore filter.
