1294706 Preparation of substituted penicillanic acid salts VSESOJUZNY NAUCHNOISSLEDOVATELSKY INSTITUT ANTIBIOTIKOV 14 Oct 1970 48906/70 Heading C2A A method of preparing 6-(3-phenyl-5-methylisoxazole-4-carbamido)-penicillanic acid sodium salt monohydrate (oxacilline) comprises reacting sodium acetate, which may be anhydrous or a 30-35% aqueous solution, with 6-(3-phenyl-5-methyl-isoxazole- 4-carbamido)-penicillanic acid. The acid may be prepared by condensation of 6-aminopenicillanic acid with 3-phenyl-5-methylisoxazole-4-carboxylic. acid chloride in the presence of a binder for hydrogen chloride, e.g. sodium bicarbonate. The condensation may be carried out in an aqueous acetone medium and the reaction product may be extracted with an organic solvent. The organic extract is dried to a moisture content of 0À8 to 1.0% as determined by the Fisher method. Pharmaceutical compositions for peroral or parenteral administration may be prepared from 6-(3-phenyl-5- methylinoxazole-4-carbamido)-penicillanic acid sodium salt monohydrate as obtained by the method described above.
