1331933 Antiviral ribonucleic acids GLAXO LABORATORIES Ltd 13 Nov 1970 [17 Nov 1969] 56169/69 Heading C3H [Also in Division A5] A process for the production of one or both of the antiviral ribonucleic acids I and II from P. chrysogenum having the characteristics: ribonucleic acid IS 20,w (as herein defined) = 11À7 to 12À3 Base analysis: Guanine 25À0% Œ 1À0 Adenine 25À0% Œ 1À0. Cytosine 25À0% Œ 1À0. Uracil 25À0% Œ 1À0. Ribonucleic acid IIS 20,w (as herein defined) = 6-7 to 7À3 Base analysis: Guanine 18À0% Œ 1À0. Adenine 32À0% Œ 1À0. Cytosine 18À0% Œ 1À0. Uracil 32À0% Œ 1À0. Said process comprising the steps of (a) culturing a strain of P. chrysogenum, preferably ATCC 10,002, 10,003, 11,707, 13,799 or a mutant thereof in or on a nutrient medium therefor whereby virus-like particles as herein defined proliferate therein; (b) rupture of the cells of the micro-organism without removing the coating of the virus-like particles, and then removal of cell debris from the virus-like particles; (c) partial or complete separation of the viruslike particles from free nucleic acids and other dissolved materials; (d) decoating the virus-like particles; and (e) fractionation whereby the desired antiviral double-stranded ribonucleic acids I and II are isolated and unwanted material is rejected. Pharmaceutical compositions may be prepared from the products.
