Abstract

1342583 1#,11α – Dihydroxy steroids and their derivatives GLAXO LABORATORIES Ltd 19 April 1971 [6 April 1970] 16255/70 Heading C2U The microbiological 1#,11α-bis-hydroxylation of ring A- and C-saturated 5α- and 5(6)-dehydropregnan-20-ones or their 20-carbonyl derivatives, e.g. ketals, possessing at the 16- and 17α- positions hydrogen atoms or only one of the following groupings viz. a 17α-acyloxy group, a 16-aliphatic group, a 16,17-epoxy group or a #16-double bond and possessing in the 3-position a keto group or a 3#-hydroxy or esterified hydroxy group, is effected using one of the microorganisms Aspergillus ochraceus, Aspergillus nidulans, Rhizopus arrhizus, Corticium praticola, or Polystictus sanguineus. The product may be recovered as an ortho-ester or alkylene dioxy derivative by reaction with for example a trialkyl o-ester of an alkanoic acid or by reaction with an aldehyde or ketone. Such derivatives may subsequently be hydrolysed to the free diols which may be acylated. The protected diols containing a 3#-OH group may be oxidized to the 3-ones or the free triols may be oxidized in the presence of a ketone to give 1#,11α-alkylenedioxy-3-ones. 3 – Keto – steroid products may be dehydrated to the corresponding #1-compounds. 3- Keto – 1#,11α – protected compounds may be converted to #4– or #4,16-derivatives by bromination and dehydrobromination. Acid hydrolysis of a 3 – keto – #4 – 1#,11α – alkylenedioxy or o-ester derivative yields a 3-keto-11α-hydroxy- #1‘4-compound. Novel products are ring A- and C-saturated 1#,11α dihydroxy – 5α – pregnane- 3,20-diones possessing at the 16- and 17α- positions H atoms or only one of the following groupings viz. 17α-acyloxy group, a 16-aliphatic group, a 16,17-epoxy group or a 16,17-double bond; the corresponding 4(5)-dehydro and 5(6)-dehydro derivatives; the 3-hydroxy and 3-esterified hydroxy steroids corresponding to both the aforementioned categories except the #4-steroids; and the 1#,11α-alkylidenedioxy, -aralkylidenedioxy and -orthoester derivatives of all the aforementioned steroids.
