1342753 Diazacyclic compounds RICHARDSON-MERRELL SpA 6 July 1971 [20 July 1970] 31754/71 Heading C2C Novel diazacyclic compounds of the general formula wherein each of R and R 1 is a hydrogen or halogen atom or a trifluoromethyl, C 1-6 alkyl, phenyl, pyridyl, C 1-6 alkoxy, hydroxy, nitro, amino, mono- or di-C 1-6 alkylamino, C 2-7 alkanoylamino, C 2-7 alkoxycarbonyl, carboxy, carbamoyl, sulphamoyl or nitrile group or R and R 1 , when at adjacent positions, may together form a methylenedioxy group, R 2 is a hydrogen atom or a C 1-6 alkyl, phenyl, pyridyl, C 2-7 alkoxycarbonyl, carboxy, carbamoyl or sulphamoyl group, R 3 is a hydrogen atom or a C 1-6 alkyl, phenyl (optionally substituted by halogen atoms or C 1-6 alkyl or C 1-6 alkoxy groups), picolyl, benzyl (optionally substituted by halogen atoms or C 1-6 alkyl or C 1-6 alkoxy groups), alkanoyl, aroyl, cyanoethyl, C 2-7 alkoxycarbonyl, formyl, carboxy, carbethoxyethyl, N – monoalkyl-, N,N – dialkyl. or N- aryl – carbamoylmethyl, N – monoalkyl or N,N – dialkyl – carbamoylethyl, C 3-6 cycloalkyl or methyl-C 3-6 cycloalkyl group, n is 1, 2 or 3, each of A and B is a -Cmq-, -Cmq-Cwx- or -Cmq-Cwx-Cyzgroup, in which each of m, q, w, x, y and z is a hydrogen or halogen atom or a trifluoromethyl, C 1-6 alkyl, C 1-6 alkoxycarbonyl, carboxy, carbamoyl, sulphamoyl, phenyl, pyridyl, benzyl or picolyl group, or m and q, w and x or y and z together form an oxygen atom, and j and k are each 0 or 1, and pharmaceutically acceptable, non-toxic acid-addition salts thereof are prepared (a) by reaction of a ketone and a heterocyclic compound of the respective general formulµ and (b) when R 3 is an alkyl or acyl group, by alkylation or acylation of the corresponding compound in which R 3 is a hydrogen atom; followed optionally by salifaction of the product. Pharmaceutical compositions having analgesic, anti-inflammatory, antipyretic, sedative, myorelaxant, anti-hypertensive, diuretic and chloretic activity comprise, as active ingredient, a diazacyclic compound of the first general formula above or a pharmaceutically acceptable, acid-addition salt thereof, together with a carrier.
