1377762 Substituted ceph-3-em-4-carboxylic acids CIBA-GEIGY AG 27 Oct 1971 [27 Oct 1970 4 March 1971 24 May 1971] 49962/71 Heading C2C Novel 7 – amino – ceph – 3 – em – 4 – carboxylic acid compounds of the formula wherein R 1 a represents hydrogen or an amino protective group R 1 A, R 1 b represents hydrogen or an acyl group or R 1 A and R 1 b together represent a bivalent amino protective group and R2 represents hydrogen or an organic radical R 2 A which together with the -C(=O)-O- grouping forms a protected carboxyl group and salts of such compounds containing salt forming groups, are prepared by oxidizing azetidin-2-one compounds wherein R 1 A, R 1 b and R 2 A have the above significance and each of R a , R b and R c represents an unsubstituted or substituted hydrocarbon group with an organic sulphoxide in the presence of an agent having dehydrating to oxidize the carbinol group to a formyl group followed by ring closure. Conversion to acid addition, organic amine quaternary, ammonium and metal salts may be carried out by normal methods using products containing a basic group at the 7-position or a free carboxylic group at the 4-position. In the case where a basic group is at the 7-position and a free carboxylic group is at the 4-position, an internal salt may be formed. In the above process it is possible, where necessary, to protect temporarily groups which do not participate in the reaction, e.g. OH, SH and NH 2 groups by acylation, tritylation or silylation and COOH groups by esterfication including silylation. Novel compounds of the first formula above and prepared by the above process may subsequently be modified to form other compounds within the said formula. Thus the examples describe the preparation of compounds by converting a 4- carboxylic ester group to the 4-carboxylic group, a 7#-acylamino group to the 7#-amino group, a 4-carboxylic group to a 4-carboxylic ester group. The starting materials for the above processes are prepared as disclosed in Specification 1,361,199. Pharmaceutical compositions having antibiotic activity contain the novel compounds of the first formula above and a suitable carrier and may be in a form suitable for enteral, parenteral or topical administration.
