Abstract

Electro-phoretically deposited components formed on a membrane from a complex substance, are individually separated into pure components by immersing the membrane in a buffer solution and eluting the components electro phoretically which are drawn off into individual collecting zones. The coated membrane 10 of accylamide gel is first made by depositing the components from the complex mixture by applying the electric field across the face of the membrane, whereby the individual components are deposited in parallel zones or “streaks”. The coated membrane is mounted in a cell 14, Fig. 1. with electrodes 18, 20, the face of the membrane being pressed against a block 22 honeycombed with channels 24 Figs 1, 5, having vertical drain grooves 26. Sponge blocks 32, 34 press the membrane 10, and a second membrane 30 against the block 22. The eluted components are drawn off from their locations in the coated membrane and drain off downwards into individual pots 28. The barrier membrane 30 prevents components from passing through, and drain grooves facing this membrane (similar to grooves 26) provide some convection flow of the buffer eluant. In modifications Fig. 7 (not shown), the pressure sponge blocks 32, 34 are replaced by bags of cellulose material which enclose the electrodes and press against the grid block 22. The buffer eluant is pumped continuously so that anolyte and catholyte are mixed neutralizing any pH change. In Fig. 9, (not shown), the pressure exerted on the coated diaphragm 10 is sufficient to cause the gel body to be forced into the channels 24. The electrodes are frames of insulating material into the edges of which perforated or wire mesh stainless steel is embedded.
