Abstract

1352104 Bleomycinic acid ZAIDAN HOJIN BISEIBUTSU KAGAKU KENKYU KAI 15 May 1972 [15 May 1971] 22726/72 Heading C2A The invention comprises bleomycinic acid of Formula I Bleomycinic acid is characterized by a melting point of 228-230‹ C. (decomposition) and an elemental analysis of C: 40À80%, H: 5À29%, N: 16À45%, O: 24À78%, S: 4À53%, Cl: 3À37% and Cu: 4À78%, is soluble in water, slightly soluble in methanol, acetic acid and dimethylsulphoxide, and insoluble in ethanol, acetic acid, acetone and ether, and gives a positive result in the Pauly and Ehrlich reactions but gives a negative result in the ninhydrin, Sakaguchi, Dragendorf, Tollens, ferric chloride, Fehling and Molisch reactions, and has ultra-violet absorption maxima at 246 mÁ and 292 mÁ and has infra-red absorption bands at 3350, 1720, 1670, 1640, 1580, 1460, 1365, 1050 and 770 cm.-1 and is hydrolysed to give 21-(2-aminoethyl)-2, 41 – bithiazole – 4 – carboxylic acid, L – threonine, 4 – amino – 3 – hydroxy – 2 – methyl – nvaleric acid, #-hydroxy-histidine, #-amino-#-(4- amino – 6 – carboxy – 5 – methylpyrimidine – 2- yl)-propionic acid, L-#-amino-alanine, L-gulose and 3-O-carbamoyl-D-mannose. A process for preparing bleomycinic comprises hydrolysing bleomycin in the presence of a strain of Fusarium genus or Helminthosporium genus or an enzyme system thereof. Suitable strains include Fusarium roseum Link emera Synder et Hansen (ATCC 20352) or (ATCC 20355) or Fusarium anguioides Sherbakoff (ATCC 20351) and Helminthosporium zonatum Ikate et Yoshida (ATCC 20353) or (ATCC 20354). The hydrolysis of bleomycin may be carried out in a phosphate buffer solution containing 0À1-0À5% bleomycin at 25‹ to 40‹ C. at pH 7À0 to 8À0. The invention also comprises a process for preparing a bleomycin which comprises hydrolysing bleomycin by a process as described above and reacting the bleomyoinic acid so produced with an amine.
