Abstract

1442477 2 – Iminomethyl – 31,41,51 – trimethoxycinnamic acid and C 1-4 alkyl esters thereof GEA AS 6 Sept 1974 [10 Sept 1973] 38983/74 Heading C2C Novel 2-iminomethyl-31,41,51-trimethoxycinnamic acid and C 1-4 alkyl esters thereof are prepared by reducing 3,4,5-trimethoxybenzylidene-cyanoacetic acid or a C 1-4 alkyl ester thereof in a neutral or alkaline medium at room temperature. The novel compounds may be reacted with guanidine to form 2,4-diamino-6-hydroxy-5- (31,41,51-trimethoxybenzyl)pyrimidine, which can be converted to 2,4-diamino-5-(31,41,51-trimethoxybenzyl)pyrimidine(trimethoprim) by treatment with POCl 3 , followed by (a) catalytic hydrogenation of the resulting 2,4-diamino-6- chloro – 5 – (31,41,51 – trimethoxybenzyl)pyrimidine, or (b) reaction of the resulting compound with hydrazine and treatment of the resulting 2,4- diamino – 6 – hydrazino – 5 – (31,41,51 – trimethoxybenzyl)pyrimidine with aqueous CuSO 4 .
