Abstract

1444947 7 – # – (Acylamino) – 3 – formyl – 3- cephem-4-carboxylic acids and cyclic isomers thereof TEKADE YAKUHIN KOGYO KK 10 Dec 1973 [8 Dec 1972 25 Dec 1972] 57179/73 Heading C2C Novel compounds of the Formulae I or III wherein R1 is hydrogen or alkoxy and R2 is acylamino or a cyclic imide group subject to the proviso that when R1 is a hydrogen atom, R2 is an acylamino group which is other than a 2- thienyl acetylamino group; and alkoxy and acyloxy derivatives of the compounds of the Formula I may be prepared by reacting a compound of the Formula II or a salt thereof with a hexavalent chromium compound optionally followed by etherification and esterification of the free hydroxyl group on the compound I. The intermediates II is prepared by (1) hydrolysis of a 3-acetoxymethyl group optionally followed by salt formation and/or (2) acylation with p-(t-butyl)-benzene sulphonyl chloride or phenyl acetyl chloride of cephalosporin C or its deacetyl derivative. N – isobornyloxycarbonyl – deacetylcephalosporin C disodium is prepared by reaction of cephalosporin C monosodium with isobornylchlorocarbonate in basic conditions. 7 – # – [D – α – ( # – methylsulphonylethoxycarbonyl)aminophenylacetamido]- 3 – hydroxymethyl – 3 – cephem – 4 – carboxylic acid sodium is prepared by acylation of 7-#-[D-α-aminophenylacetamido) – 3 – acetoxoxymethyl – 3- cephem – 4 – carboxylic acid with # – methylsulphonylethoxycarbonyl chloride followed by treatment of the free acid with sodium bicarbonate. Pharmaceutical compositions of the compounds I and III have antimicrobial and bacterocidal activity.
