Abstract

1,259,321. Insolubilized enzymes. CORNING GLASS WORKS. 5 Feb., 1969 [5 Feb., 1968], No. 6144/69. Heading C3H. [Also in Division C1] A stabilized enzyme composition which is substantially water-insoluble and capable of repeatedly catalysing reactions without loss of activity comprises an inorganic carrier having a surface area of greater than 0À1 m2/g. which is a porous glass, a colloidal silica, wollastonite or a porous dried silica gel, said carrier having many uncontaminated reactive silanol groups on such surface that are ionically bonded to the enzyme by amine-silicate bonds and that are hydrogen-bonded to functional groups on the enzyme, to provide an enzyme composition of substantially constant activity. The porous glass may be as particles, or as a disc or cylinder. The enzyme may be papain, ficin, pepsin, trypsin, chymotrypsin, bromelin, keratinase, cellulase, amylase, maltase, pectinase, chitinase, lipase, cholinesterase, lecithinase, phosphatase, ribonuclease, desoxyribonuclease, arginase, asparaginase, glutaminase, urease, glucose oxidase, catalase, peroxidase, lipoxidase, a cytochrome, glutamic-pyruvic transaminase, glutamic-oxalacetic transaminase, transmethylase or phosphopyruvic transphosphorylase. The bonding may be brought about by allowing a buffered aqueous solution of the enzyme to react with the carrier, which may first be cleared, at below room temperature.
