Abstract

1383874 À Antibiotics UPJOHN CO 10 Sept 1973 [24 Oct 1972] 42366/73 Heading C2A New antibiotics EM-2, EM-3 and EM-4 are obtained by the fermentation of the microorganism Emericellopsis microspora strain 333, NRRL 5648 in an aqueous nutrient medium to which propylproline is added. Antibiotic EM-2 (a) has the following elemental analysis: C, 57À47; H, 7À70; N, 12À99; O, 21À87, (b) has a M.Pt. of 261À4 C.; (c) has a specific rotation [α]25 D = +5 degrees; (d) has the following U.V. spectrum in methanol: (e) is soluble in dimethylformamide, dimethyl. sulphoxide and C 1-4 alcohols; is less soluble in chlorinated hydrocarbon solvents and ethyl acetate; is rather insoluble in acetone and ether; and is completely insoluble in saturated hydrocarbon solvents; and (f) has the I.R. spectrum indicated in Fig. 1 (not shown). Antibiotic EM-3 (a) has the following elemental analysis: C, 58À46; H, 7À78; N, 13À27; O, 21À49; (b) has a M.Pt. of 256À9 C.; (c) has a specific rotation [α]25 D = + 12 degrees; (d) has the following U.V. spectrum in methanol; (e) has the same solubility characteristics as EM-2; and (f) has the I.R. spectrum indicated in Fig. 2 (not shown). Antibiotic EM-4 (a) has the following elemental analysis: C, 57À26; H, 7À45; N, 14À22; O, 21À08; (b) has a M.Pt. of 239À8 C.; (c) has a specific rotation [α]25 D = + 13À5 degrees, (d) has the following U.V. spectrum in methanol: (e) has the same solubility characteristics as EM- 2; and (f)has the I.R. spectrum indicated in Fig. 3 (not shown). The antibiotics EM-2, EM-3 and EM-4 are isolated from the fermentation broth and are separated into their individual entities by silica gel chromatography.
