Abstract
1392551 Ergolene derivatives SANDOZ Ltd 16 May 1972 [19 May 1971 (3) 15 Dec 1971] 22874/72 Heading C2C Novel compounds of the Formula (I) wherein each of R 1 and R 2 is H, or C 1-4 alkyl or R 1 and R 2 together form an alkylene chain of at most 5 carbon atoms, may be obtained by (a) reacting an amine of formula- HNR 1 R 2 with an acid azide of Formula (II) in an aprotic solvent after the compound of Formula II has been caused to rearrange in known manner with elimination of nitrogen to form the isocyanate, or (b) N-carbamoylating the -NH 2 group in a compound of Formula (V) with a reagent known to be capable of introducing the group -CONR 1 R 2 . Novel compound (V) 6-methyl-8#-aminomethyl-ergolene is obtained by the steps of treating lysergol with methane-sulphonic acid chloride to form 6-methyl-8#-mesyloxymethyl-ergolene which is reacted with alkali azide to form 6-methyl-8#-azidomethyl-ergolene, and the latter is reduced to form compound (V). The acid azide (II) is obtained by converting 6-methyl-9-ergolene-8-#-acetic acid to a reactive derivative, e.g. the acid chloride or bromide, and reacting with an alkali azide. Pharmaceutical compositions having antihypertensive properties comprise compounds of Formula I, in free base or acid addition salt form, in association with a pharmaceutical carrier or diluent, suitably in unit dosage form or in sustained release form.
