Abstract
The device for delayed pharmaceutical release is used in an aqueous environment in the human or animal body. It essentially releases the pharmaceutical by diffusion. It displays an integral solid structure of such a shape and size that it can be introduced into the environment in which it is to be used and can be held there. It consists of a mixture which contains a non-ionic pharmaceutical, a polymer and, in addition, 0.1 to 15% by weight of a soluble substance with osmotic activity. The pharmaceutical has a solubility in water of from 200 to 10,000 ppm. The polymer is permeable to water and to the pharmaceutical, is insoluble in the aqueous environment at least for the duration of the pharmaceutical release and is impermeable to the soluble substance. The soluble substance is in the form of single depots which have a number average diameter of 1 to 250 mu m which are dispersed in the polymer so that they are surrounded essentially singly by a polymer layer. When the device is introduced into the aqueous environment, the depots absorb water and rupture the surrounding polymer layers. This results in a network of mutually connected pores inside the device. There is thus an in situ enlargement of the surface area on which the pharmaceutical can be released from the polymer to the environment. This in situ enlargement of the surface area increases the rate of release of the pharmaceutical from the device.
